Plot summary
The singer at Gus Jordan's Bowery saloon is Lady Lou, and her boss keeps her supplied with diamonds. She has more male friends than anyone might imagine. Dan Flynn, a rival of Jordan's, drops hints that the saloon's business is crooked and offers to look after Lou once Jordan lands in jail. Jordan makes his money from prostitution and from counterfeiting, and he ships young women to San Francisco, where they work as pickpockets. The mission next door is run by Captain Cummings, and his efforts to clean up the neighborhood worry Jordan because they could scare off his customers. Lou's former sweetheart, Chick Clark, is in prison for a robbery he carried out to get diamonds for her, and she is warned that he believes she has betrayed him.

- The Hays Code declared Mae West's play Diamond Lil banned from the screen and demanded the film drop elements of it; the adaptation was allowed only if the play was not mentioned in publicity, and Joseph Breen blocked re-releases in 1935 and 1949.
- She Done Him Wrong was nominated for the Academy Award for Outstanding Production, now known as Best Picture, and at 66 minutes it is the shortest film ever to be so honored.
- Mae West personally brought Louise Beavers, the only African-American actress in She Done Him Wrong, into the film, and made a point of acting with Black performers on stage and screen, helping to break racial discrimination in entertainment.
- Mae West always claimed she discovered Cary Grant for She Done Him Wrong, saying he had only made "some tests with starlets" before, though Marlene Dietrich's Blonde Venus and Sylvia Sidney's Madame Butterfly both came out earlier.
- Rafaela Ottiano, who played Russian Rita in the Broadway version, was cast as Russian Rita in the film on Mae West's recommendation, which led to work with West throughout her career.
