Plot summary
Alice and her clones storm Umbrella's Tokyo headquarters, cutting down the branch until Albert Wesker escapes by plane and sets off a bomb. The real Alice, aboard, tries to kill him, but he injects her with an anti-virus that strips away her superhuman powers. The plane crashes in the mountains, and Alice walks away. Six months later she flies to Alaska, following broadcasts from Arcadia, a supposed safe haven. She finds only a field of abandoned planes, where Claire Redfield attacks her, a scarab device fixed to Claire's chest. Alice destroys the device, which calms Claire but leaves her memory gone. The two fly to Los Angeles and find survivors holding a prison ringed by thousands of the undead, led by Luther West.

- The size and weight of the 3D cameras meant the filmmakers could not use existing equipment such as Steadicam rigs, so the camera operator worked from a Segway to get the look of a Steadicam shot.
- Toronto police received hundreds of calls from concerned neighbors after the production filmed a scene involving a burning plane crash.
- Anderson was shown footage of James Cameron's Avatar, which sold him on shooting Resident Evil: Afterlife in digital 3D using Cameron's Fusion Camera System and a Sony F35. The crew spent two weeks in pre-production simply learning the 3D camera system.
- Filming in 3D added 20% to the budget, and during production Jovovich accidentally shot out a $100,000 camera.
- Johnny Messner auditioned for Chris Redfield before Wentworth Miller was cast. Having never played the Resident Evil games, Miller prepared by searching the internet for images of the character and was surprised how muscular he was, knowing that with three weeks to prep he could not match it.
