Plot summary
James McCullen's company M.A.R.S. builds nanotech warheads that consume any non-organic material, and sells four of them to NATO. American soldiers Duke and Ripcord are assigned to deliver the shipment. On the road their convoy is ambushed by thieves under the command of the Baroness, and Duke recognizes her as Ana Lewis, the woman he once planned to marry. A covert international team, the G.I. Joe unit, intervenes and drives the attackers off, then takes the warheads to its command center in Egypt, known as the Pit. Its leader, General Hawk, allows Duke and Ripcord to join the unit after Duke admits that he knows the Baroness.

The Cobra Commander mask worn by Joseph Gordon-Levitt was redesigned from its comic-book look because the film crew felt the original design resembled a Ku Klux Klan hood.- While filming in Prague on April 26, several people were injured when a bus and cars collided with a four-wheel-drive vehicle involved in the shoot that appeared to have braking problems, though emergency services said the injuries were minor.
- As a marketing stunt, more than 300 parachute-equipped, twelve-inch G.I. Joe action figures were dropped from the roof of a 42-story Kansas City hotel, soaring 500 feet to the ground at the International G.I. Joe Convention.
