Plot summary
Dante Hicks and Randal Graves bought the Quick Stop store fifteen years ago, and they spend their days there. Jay and Silent Bob run a marijuana dispensary in the former RST Video. Dante is still grieving Becky and their unborn daughter Grace, who were killed by a drunk driver. Randal has a severe heart attack at work. After surgery, his doctor asks Dante to keep Randal in good spirits and warns that Dante faces the same risk. Randal declares his life wasted and says he will make a movie about the store where he and Dante work.

- Kevin Smith's 2018 near-fatal heart attack led him to rewrite the Clerks III script from scratch and discard the original storyline; he later said the old draft strayed too far from the original Clerks and was written by "a guy who didn't know a thing about death," and that he was glad it was never made.
- Plans for Clerks III came to a halt in 2017 when one of its four leads, later revealed by Kevin Smith to be Jeff Anderson, opted out of reprising his role despite a completed script; Smith doubted at the time that the film would ever be made.
- Kevin Smith said an ongoing audit over Clerks II residuals with The Weinstein Company was delaying key Clerks III cast and crew, including Jeff Anderson and Scott Mosier, from coming on board until it was resolved.
- Kevin Smith called his film Tusk "the absolute bridge" to Clerks III, saying that because of Tusk he got his financing for Clerks III.
