Plot summary
Scat Sweeney and Hot Lips Barton play clarinet and trumpet in a swing quintet until the group breaks up. They drift across the country looking for work, moving on whenever a woman complicates things, and end up stowing away on a ship headed for Rio after a circus fire. On board they cross paths with Lucia, a nightclub singer who is distraught, thanks them for their help, then hands them over to the captain. Lucia is under the hypnosis of her guardian, Catherine Vail, who intends to marry her off to her brother once the ship docks, a match that would give Vail control of Lucia and of some important papers.

- Bing Crosby and Bob Hope bought a substantial interest in Lime Cola and, as majority owners of Road to Rio, insisted that signs and posters for the drink appear in the background throughout the film; Paramount president Y. Frank Freeman objected, but the posters stayed in.
- The Lime Cola plugs came to nothing: demand for the drink far exceeded supply, the company filed for reorganization in April 1948, and Crosby and Hope, who held heavy interests in it, lost their considerable investments.
- For a song promoting Lime Cola, Crosby and Hope wanted a city name containing "cola"; Florida's Pensacola was too close to rival Pepsi-Cola, so they settled on Apalachicola and chanted "Apalachi-COLA!" in the film's first song number.
