Plot summary
Five years after the T-virus escaped Raccoon City, it has spread across the globe and stripped the land of life. Alice wanders the wasteland alone, fights off marauders, and finds a notebook that points to a supposedly uninfected region in Alaska. A convoy of survivors roams the desert for supplies and safe harbor, led by Claire Redfield with Carlos Oliveira and L.J. Wade. Below ground, Umbrella's Dr. Isaacs works on an anti-virus to tame the infected, growing clones of Alice to further his research, while the corporation's chairman, Albert Wesker, has him watched. When infected crows fall on the convoy, Alice uses a new power to drive them off.

Can contain spoilers, watch the movie first.
- Sienna Guillory, who played Jill Valentine in Resident Evil: Apocalypse, was planned to return as Jill in Extinction but passed on the part because of her commitments to the film Eragon.
- Claire Redfield did not exist in the early drafts of Extinction; the plan then was for Jill Valentine, played by Sienna Guillory in Apocalypse, to carry on. Producers Paul W. S. Anderson and Jeremy Bolt instead chose to put Alice alongside a different game heroine.
- The shoot was moved from the Australian Outback to Mexicali, Baja California, Mexico, where principal photography began in May 2006. The change of locations pushed the production dates back repeatedly, from November 2005 to May 2006.
- Extinction was the first film in the series to show herbs, the healing item from the video games, in Dr. Isaacs' lab. It also reused the first film's Hive replica, the Raccoon City Hospital, the White Queen computer and the Spencer Mansion replica.
