Plot summary
At the San Diego Zoo Safari Park, primatologist Davis Okoye is friends with George, a leucistic western lowland gorilla he once rescued from poachers, and the two communicate in sign language. A canister of a pathogen lands in George's habitat, and the gorilla grows much larger and turns aggressive. Genetic engineer Kate Caldwell then contacts Davis and explains that Energyne created the pathogen to rewrite genes on a large scale, and that she had hoped her research would lead to cures for diseases before she learned the company intended it as a weapon. Canisters have also reached a crocodile named Lizzie in the Everglades and a wolf named Ralph in a Wyoming forest. Davis and Kate set out to find a way to undo the change.

- In late March 2018 German director Uwe Boll threatened to sue Warner Bros. unless it changed the film's title, saying it would shrink his brand and the revenues he could use for future installments of his own unrelated Rampage trilogy, about a spree-killing revolutionary.
- Rampage was the last released Warner Bros. film financed by Access Entertainment / RatPac-Dune Entertainment; the studio ended the partnership over sexual harassment allegations against owner Brett Ratner, leaving the company uncredited, and it dissolved later that year.
- Rampage became the best-reviewed live-action video game film in Rotten Tomatoes' history, a record it held until Pokémon Detective Pikachu was released the following year.
- Before the producers settled on Ryan Engle's script, writers pitched many takes, including a faithful one to the game in which the monsters were mutated humans; Engle aimed for a "love letter" to monster movies such as Jaws and Jurassic Park.
- Weta Digital's effects supervisor Erik Winquist and a small crew traveled to Chicago to create a model of the Chicago Loop for the climactic battle, taking up to 15,000 photographs with 3D scanners while motion cameras covered downtown Chicago.
- To stage the destruction of Chicago's buildings, Weta's artists studied the World Trade Center collapse of 2001 and the implosions of buildings affected by the 2016 Kaikōura earthquake in their hometown of Wellington.
- Motion capture coach Terry Notary took a break from Avengers: Infinity War, which was also filming in Atlanta, to help Jason Liles with his performance as the gorilla George.
