Plot summary
Murders have struck New York City over several months, always at night beneath a full moon, with each victim's body cannibalized afterward. Witnesses speak of a disfigured monster. Because the scalpel used on the victims belongs only to Dr. Xavier's medical academy, the police suspect its staff, and they give Xavier 48 hours to find the killer himself. Wells, Haines, Duke and Rowitz fall under suspicion. Reporter Lee Taylor pursues the story and meets Xavier's daughter Joanne, who is cold to him.

- While making Doctor X, the studio also shot an alternative black-and-white version that still exists; side-by-side comparison shows most takes match, differing only in small moments such as Tracy's ad-lib with a skeleton in a closet and Mae Busch's dialogue as a brothel madam.
- The Rocky Horror Show and its film adaptation open with "Science Fiction/Double Feature", which name-checks classic horror films, among the references is the line "Doctor X will build a creature", even though Doctor X never builds a creature in the film.
- Progressive metal band Queensrÿche paid homage to the film by featuring a character named Doctor X, called Dr. X in the lyrics, as the main antagonist of their 1988 concept album Operation: Mindcrime.
- A line of dialogue from early in Doctor X was sampled on the album Dr. Octagonecologyst by rapper Kool Keith and producer Dan the Automator.
- Doctor X was one of the last films shot in the early two-color Technicolor process, and its release was handled unusually: black-and-white prints were shipped to small towns and foreign markets, while color prints were reserved for major cities.
