Plot summary
Worthington Labs announces an inoculation that suppresses the X-gene, the source of mutant abilities, and says any mutant may take it. Rogue's touch harms anyone she touches, so the offer interests her, while other mutants are wary. Magneto gathers those who oppose the cure into his Brotherhood of Mutants and warns that it will be used as a weapon against his kind. Hank McCoy, a former student of Xavier's school, now serves in the U.S. Cabinet as the Secretary of Mutant Affairs.

- Channing Tatum was in the running for the role of Gambit, but the character was written out of the film.
- Maggie Grace was considered for the role of Kitty Pryde before Ratner invited Elliot Page, who had impressed the director with his performance in Hard Candy. Page initially declined, not wanting to jump to Hollywood filmmaking yet, and accepted only after reading the script.
- An early treatment focused on Jean Grey's resurrection and would also have introduced the villainess Emma Frost, an empath manipulating Jean's emotions, in a role intended for Sigourney Weaver.
- Matthew Vaughn, signed to direct and already casting Kelsey Grammer as Beast and Vinnie Jones as Juggernaut, withdrew before filming began. In October 2023 he said he left after a group of executives had tried to sign Halle Berry on with a fake script, which included scenes of Storm rescuing kids from Africa.
- In 2017, during the MeToo and Time's Up movements, Elliot Page accused Ratner of homophobia, saying the director outed him as a lesbian at a cast and crew meet and greet during production, which made him feel violated. Anna Paquin said she was present when Ratner made the comment.
- A notable effect was the “digital skin-grafting” that rejuvenated the faces of senior actors Patrick Stewart and Ian McKellen, made by the Brothers Strause's Lola Visual Effects.
- San Francisco vetted any filming on the real Golden Gate Bridge, including aerial shooting, so the effects team spent two months on a miniature in Santa Clarita, shooting one minute of footage a day at “golden hour”, complete with explosives.
- The $210 million budget made The Last Stand the most expensive film to be made at the time, a record first broken by Pirates of the Caribbean: Dead Man's Chest (2006) and its $225 million budget.
