Plot summary
In the wake of a vampirism pandemic, America's towns and cities lie abandoned, and survivors crowd into rural pockets, dreading nightfall. Martin's family is killed, and a hunter known only as Mister takes the teenager along, teaching him to fight the feral vampires as they travel north through locked-down heartland towns toward New Eden, the haven said to survive in Canada. On the road they are joined by Sister, a nun Mister rescues from two men. To avoid The Brotherhood, a fundamentalist militia whose leader, Jebedia Loven, preaches that the plague is God's will, they keep to the back roads.

- Director Jim Mickle and co-writer Nick Damici first conceived Stake Land as a cheap weekend web series and wrote forty 8-minute scripts for it, before producer Larry Fessenden convinced them to turn it into a feature film instead.
- The scene where the Brotherhood drops vampires into a survivors' settlement was never meant to be one continuous shot; director Jim Mickle only filmed it as a single take because scheduling limits forced the whole sequence into a single day, and afterward only a few frames were trimmed to remove poor acting from extras.
- Belle was originally written as an older character and something of a love interest for Mister, but after interviewing Danielle Harris, Nick Damici thought she seemed more like his character's "grandkid", and they rewrote the script accordingly.
- Kelly McGillis, who plays the nun Sister, took the role after a ten-year break from acting; Stake Land marked her return to feature films.
