Plot summary
At the United Fraternity national convention in Fort Lauderdale, the Adams College chapter of Lambda Lambda Lambda has booked rooms at the Royal Flamingo Hotel. The acting manager, Buzz, has handed those rooms to the Alpha Betas instead, and receptionist Sunny Carstairs cannot talk him out of it. Poindexter finds the group other rooms at the Hotel Coral Essex, a rundown property in a rough neighborhood. The Alpha Beta president, Roger Latimer, with Ogre from Adams College at his side, sets out to push the Tri-Lambs out of the convention. He gives them wrong directions to a pre-convention barbecue and leads them into a trap, where disguised Alpha Betas force them to strip to their underwear and chase them off.

- Curtis Armstrong, who played Booger, hated the Revenge of the Nerds II script and turned down repeated offers with pay raises, preferring to focus on Moonlighting, despite pleas from co-star Robert Carradine. He accepted after a Fox executive told him Larry Gelbart would serve as script doctor, only to learn Gelbart had never been approached.
- Julia Montgomery, who played Betty Childs, was set to appear in Nerds II but refused after learning her only scene had Betty cheating on Lewis in a hotel room with another jock; she felt that was wrong after Betty fell in love with Lewis in the first film.
- Anthony Edwards disliked the script and was reluctant to return as Gilbert, agreeing only when his scenes were scheduled to shoot very fast and with no location filming. The script was rewritten to give Gilbert a broken leg so Edwards could film almost all of his material in one session.
- Brian Tochi could not return as Toshiro Takashi in Nerds II because he had already contractually agreed to play Tomoko Nogata in the Police Academy series, a deal he signed before learning the Nerds sequel was being made.
- The sequel was originally to be set in Puerto Vallarta, Mexico, but was moved to Fort Lauderdale, Florida because that was a faster and cheaper alternative. Shooting there during spring break let the production use vacationing college students as extras and background actors.
