Plot summary
Carl Peterson and Molly Thompson marry in Hawaii, where Dupree, Carl's best man and longtime friend, nearly misses the ceremony after landing on the wrong island. Back home, Molly's father Bob, the CEO of the company Carl works for, promotes Carl and then presses him with odd conditions, among them a vasectomy. Dupree, meanwhile, has lost his job, his car and his home, and Carl and Molly let him sleep on their couch. He is a messy guest, he befriends the neighborhood children and takes a librarian out, while Carl grows uneasy about how comfortably Dupree settles into the house. Molly's patience with their guest wears thin.

Can contain spoilers, watch the movie first.
- The film's title caused a stir because Dupree shares its name with the title character of Steely Dan's song "Cousin Dupree". Founders Donald Fagen and Walter Becker wrote a tongue-in-cheek letter to Luke Wilson about his brother Owen Wilson's "appropriation" of the name, inviting him on stage to apologize to fans.
- Composer Rolfe Kent scored You, Me and Dupree, but at the very last minute, a week before the press screenings, his score was thrown out and replaced by one written by Theodore Shapiro.
- The scene where Dupree arrives by light plane on the wrong island was shot at Kualoa Ranch, the same valley in which Jurassic Park was filmed.
