Plot summary
Amanda Waller runs the black-ops Task Force X program, and she fills its roster from Belle Reve prison, offering inmates commuted sentences. Harley Quinn, Peacemaker, King Shark, Polka-Dot Man and Ratcatcher 2, who brings her pet rat Sebastian, are among the recruits; Bloodsport joins after Waller threatens to send his daughter Tyla to prison, and Rick Flag serves as field leader. An anti-American regime has taken power on the island nation of Corto Maltese, and the squad is sent there to destroy Jötunheim, a Nazi-era laboratory housing the secret experiment Project Starfish. The squad lands on an island held by the regime's military, with a rebel faction under Sol Soria also fighting there.

- Warner Bros. hired James Gunn to write the next Suicide Squad film after Disney and Marvel Studios dismissed him as director of Guardians of the Galaxy Vol. 3 in July 2018, when commentators including Mike Cernovich circulated controversial Twitter posts he had made.
- Gavin O'Connor wrote three-quarters of a screenplay for the film centered on a father-daughter story between Deadshot and his daughter Zoe, but left after new president Walter Hamada asked him to make it a comedy and he declined.
- The film featured the biggest sets ever built for a Warner Bros. production, including a Jötunheim exterior the size of three football fields where the final battle was shot, and an even bigger beach set with real waves and a palm tree forest.
- Daniela Melchior was cast as Ratcatcher 2 from a group of 200 actors, and did a chemistry test with live rats during her audition because her character controls rats and had to interact with them on set.
- James Gunn wrote Peacemaker for Dave Bautista, who declined because of a scheduling conflict with Zack Snyder's Army of the Dead; John Cena took the part after numerous unsuccessful attempts to join the DCEU.
- James Gunn met Idris Elba on March 1 to discuss him joining the film, reportedly as a replacement for Will Smith as Deadshot; Elba was Gunn's only choice for the role and agreed during that conversation, with Gunn writing the script with him in mind.
- The film's first trailer was viewed 150 million times within a week of its release, breaking the record for red band trailers that had been set earlier in the year by the trailer for Mortal Kombat.
