Plot summary
A year after Shredder was stopped, the Turtles still live in the shadows while the public credits Vern Fenwick for the victory. April O'Neil discovers that the scientist Baxter Stockman works for Shredder and plans to free him during a prison transfer. The Foot Clan attacks the convoy, and Stockman's teleportation device sends Shredder into another dimension, where the alien warlord Krang gives him a mutagenic compound and orders him to rebuild a device that opens a portal. The mutagen turns two criminals, Bebop and Rocksteady, into a warthog and a rhinoceros. April takes the remaining vial, the Foot pursues her, and corrections officer Casey Jones comes to her aid. In the lair, Donatello learns the mutagen could make the brothers human, and Leonardo tells him to say nothing.

- It was a box-office bomb, grossing only $246 million against a $135 million budget, which resulted in a planned third film being scrapped in favour of an animated reboot, Mutant Mayhem, released in 2023.
- The Hollywood Reporter estimated that the film lost the studio at least $75 million once all expenses and revenues were factored together.
- Fred Armisen was originally chosen to voice Krang, but in May 2016, shortly before the film's release, Brad Garrett voiced Krang instead because Armisen had schedule conflicts.
- Filming began in April in New York City and Buffalo, spending about $70 million in the state; the crew also moved out to Foz do Iguaçu in Brazil, and Buffalo work ran along the Kensington Expressway from 4 May to 17 May.
- In an interview William Fichtner stated that he would be returning as Eric Sacks, but for unknown reasons he ultimately did not appear in the final film.
- Gary Anthony Williams was cast as Bebop and WWE wrestler Sheamus was confirmed to play Rocksteady; CM Punk later revealed that he had lost the role of Rocksteady to Sheamus.
