Plot summary
Susan Cooper spends her days at a CIA desk, linked remotely to her partner, the field agent Bradley Fine. Fine accidentally kills the arms dealer Tihomir Boyanov, who dies before he can reveal where a suitcase nuke is hidden. Susan also learns that Boyanov's daughter, Rayna, has contacted the terrorist middleman Sergio De Luca. Since Susan has never worked in the field, Rayna does not know her, and Susan volunteers to track Rayna herself. Her boss, Elaine Crocker, agrees only reluctantly, and agent Rick Ford quits over her inexperience. Susan's best friend and colleague, Nancy, supplies intelligence from afar as Susan leaves on her first field assignment.

- Zooey Deschanel was originally slated to play the role of Elaine Crocker but left the project; 50 Cent joined the cast playing himself, and Nia Long was cast too but did not appear in the finished film.
- A prosthetic was used for shots of Nicola (Julian Miller) exposing himself, and the production team had to present the prosthetics used in those shots to the MPAA for rating.
- Paul Feig said Spy went through about 10 test screenings, a process that includes recording the audience laughter and which he does religiously; Judd Apatow said it doesn't work very well if a movie is supposed to make you feel difficult emotions, but for comedy it is often the best way to refine jokes.
