Plot summary
In 2008, Rob Gibson coaches his Nashville high school boys' basketball team to a third straight state championship. Fifteen years on, he is going through a divorce and loses his coaching job. He takes a job at a drag queen restaurant and runs into Alex Cruise there, the point guard from that championship team. Rob begins training Alex for a track and field event with a cash prize, and when Alex is mistaken for a transgender athlete, Rob pushes him to enter the women's competition instead. Alex wins every event, and reporter Gwen Wilde notices, offering to help the pair chase fame. Rob and Alex bring the old teammates back together to play in a women's basketball league, passing themselves off as transgender athletes.

- Lady Ballers began as a pitch for a documentary in which men would pose as transgender athletes to infiltrate women's sports; The Daily Wire switched to a fictional comedy because women's leagues do not admit men. Ben Shapiro said the male actors were unwilling to undergo the procedures needed to play on women's teams.
- While filming audience scenes in Nashville, several extras objected after learning the film's political nature and the Daily Wire's involvement and were escorted out of the building. The next day a "Baller Pride" sign and a transgender flag were removed from the set, and recruiters doubled extra pay to $300 a day.
- Belmont University canceled planned filming of Lady Ballers on its campus, saying the production company had not fulfilled the requirements or provided the details that would have been necessary for the shoot to proceed.
