Plot summary
Gru has left supervillainy to raise Margo, Edith, and Agnes and run a jelly business. When a mutagen called PX-41 is stolen from an Arctic lab, the Anti-Villain League recruits him. Agent Lucy Wilde brings him in, though he refuses at first, citing his duties as a father. After his assistant Dr. Nefario retires to work for another villain, Gru agrees to help. He and Lucy pose as bakers at a shopping mall. Gru suspects Eduardo Pérez, owner of a Mexican restaurant, of being El Macho, a villain thought dead twenty years ago. Agnes hopes Gru will fall in love with Lucy, but he insists their partnership is professional. A blind date arranged by neighbor Jillian goes wrong until Lucy rescues him, and they begin to bond.

- Al Pacino was cast to voice the villain El Macho and recorded all of his dialogue for the film, but he left in May 2013 over creative differences and was replaced by Benjamin Bratt.
- Because the animation was already finished around Al Pacino's original performance, Benjamin Bratt had to match his line readings exactly to the character's existing mouth movements, and after trying and failing to imitate Pacino's voice, he gave up and used his own interpretation instead.
- Pharrell Williams’s song “Happy”, the album's only single, was released with a 24-hour music video, a first for any film, and that video's viral response helped drive the song's global success.
- Rumours claimed the film's release was banned in China because the Minions too closely resembled former Chinese president Jiang Zemin. China's Film Bureau was “furious” about the negative comments, stating that the film had not been submitted for censorship approval.
- Despite costing only $76 million to make, Despicable Me 2 became the single most profitable film in Universal's 101-year history at the time.
- Javier Bardem was in talks to voice the villain El Macho, but the negotiations fell apart and he chose to play the villain in Skyfall instead.
- An idea featuring the Minions with superpowers was cut, so the filmmakers used the evil versions of them instead, and the superpower idea was finally used years later in Despicable Me 4 in 2024.
