Plot summary
Dr. Paul Henderson, his wife Carla, and Jim Farrell arrive on a tropical island. Henderson is investigating radiation from atomic bomb tests; Farrell is a Peace Corps representative. The captain tells them the island is known as Blood Island. They witness a funeral for two dismembered bodies and meet Arcadio, the village elder, and his granddaughter Alma. Two village women are chosen for a ritual. Esteban Powers invites the Hendersons to stay at his home; he says the islanders were resettled after atomic bomb tests and the plants and animals have changed. On the way back, a banana tree grabs Carla and a butterfly bites Paul. That night, a deformed beast emerges from the jungle and mutilates the two women at the altar. Alma tells Jim the beast only wants women.

- Female patrons were promised the chance to "become a Bride of Blood" and get a free engagement ring set, two cheap plastic rings each. Sam Sherman devised the gimmick and wanted to add a phony printed marriage license, but Kane W. Lynn vetoed it, fearing legal trouble involving minors such as unwanted pregnancies.
- The shoot was scheduled to run four weeks but stretched to 11 because the money kept running out. John Ashley, who got along well with Eddie Romero and the crew, would stop working whenever his pay was late.
- John Ashley said he was keen to accept the role because his marriage to Deborah Walley had just broken up and he wanted to get out of the United States.
- The film's success led to two more "Blood Island" films, The Mad Doctor of Blood Island and Beast of Blood. John Ashley moved to the Philippines in 1968 and lived there for years, co-producing several horror films with Eddie Romero and Roger Corman.
- The last third of the film ignores day-and-night continuity, cutting from dark skies to bright blue skies and back again.
