Plot summary
Cragwich lives under a curse. When Baron Wolfgang MacLaren cut off the head of the vampire queen Carmilla, she swore that every girl in the village would turn into a vampire on her eighteenth birthday. In the present day, Jimmy is dumped by his girlfriend Judy, and Fletch is let go from the job where he dressed as a clown. The two friends set off to camp in the countryside, letting a dart thrown into a pub map choose their destination: Cragwich. At the bar Baron's Rest, the innkeeper offers them the Mircalla cottage in the woods. There they meet Lotte, Heide, Anke and Trudi, folklore students studying the legend of Carmilla. Their van breaks down, and the women give the men a ride. Jimmy and Lotte fall for each other, and Fletch hopes for beer and women.

- When Lesbian Vampire Killers came out on DVD and Blu-ray on 3 August 2009, Momentum Pictures claimed retailers, including the supermarket chain Tesco, demanded warning stickers be placed over the word "Lesbian"; Tesco said it had only asked for a cover with less cleavage.
- AV Pictures offered Phil Claydon the director's job after running into difficulty securing funding; Claydon approached James Corden after seeing him in The History Boys, and Corden then sent him a pre-release copy of Gavin & Stacey, which convinced Claydon to cast Mathew Horne as co-star.
- Claydon said of the film's special effects, "I covered James in vampire gunk at every opportunity because that made me laugh", the vampires turning into slime rather than dust or bursting into flames.
