Plot summary
Robert Campbell works alone in the Amazon rainforest, where his wife and research partner left him. Aston Laboratories, the drug company he works for, sends Dr. Rae Crane with equipment and supplies and orders to judge whether his research deserves continued funding. Campbell asked for a research assistant and is unhappy that a woman arrives instead; he tries to send her back. He believes he has found a cure for cancer but cannot synthesize the compound. A logging company is building a road toward the village.

- Director John McTiernan sought out Swedish actress Lena Olin for the female lead Dr. Rae Crane because "she was strong and she was very smart", but Sean Connery disapproved, worried that having her co-star opposite him would result in her stealing his prowess.
- McTiernan later explained that Connery's Bond-movie background left him wary of an equally commanding female lead, saying the film needed a woman "as big as" Connery himself, and that he ultimately failed to convince the star of that idea.
- Director John McTiernan later argued the film's modest reputation was undeserved, calling it "a little art movie with Sean Connery that cost only $27 million" and blaming the press for setting expectations by marketing it as an action film.
- Tom Schulman's script was purchased for $2.5 million, and the production then spent a further $1 million on rewrites by Sally Robinson and Tom Stoppard.
- Lorraine Bracco's performance in Medicine Man, combined with her role in Traces of Red, earned her a Golden Raspberry Award nomination for Worst Actress at the 13th Razzies.
- Filming took place in the Mexican jungle near Catemaco starting March 4, 1991, and both Sean Connery and Lorraine Bracco later complained about the on-set conditions there.
