Plot summary
In 1962 Baltimore, teenager Tracy Turnblad dreams of dancing on The Corny Collins Show, a local television program whose dancers are all white. Against the odds, Tracy wins a place on the program and becomes a celebrity overnight, and she meets Link, the resident dreamboat, and Amber, the ambitious mean girl. In detention she meets Seaweed, an African-American boy, and his mother, Motormouth Maybelle, who owns a local record store. Amber's mother is Velma Von Tussle, and the two of them are pro-segregation. Tracy's mother is Edna Turnblad. Live audiences appeared in some scenes as Baltimore bystanders and as the program's studio audience.

- NBC cast Tracy Turnblad through an open call in New York City, hoping to repeat the discovery of Shanice Williams in The Wiz Live!; the role went to college sophomore Maddie Baillio, her first professional role.
- Unlike NBC's earlier live musicals, Hairspray Live! was staged from the Universal Studios backlot, with much of it performed outdoors, and live audiences were used as on-stage extras, playing Baltimore bystanders and the studio audience of The Corny Collins Show.
- Two songs written for the 2007 film, "Ladies' Choice" and "Come So Far (Got So Far to Go)", were added to Hairspray Live!, while the Broadway number "The Big Dollhouse" was cut and "The New Girl In Town" was left out.
