Plot summary
Jamar and Suwo grow up in the American West after their uncle Arana carries them out of the Dutch colony as newborns. Their father, the sultan Hamza, fought foreign tyranny. In 1860 the two brothers finish a stint on the Transcontinental Railroad with Arana, and on the train home they fight off a band of gangsters. Back in Java, the brothers find a village locked in a struggle with Captain Van Trach, a Dutch administrator, and his henchmen. Arana and his nephews take up the villagers' cause, and Jamar and Suwo set out to punish the man responsible for their family's ruin.

- Yoshi Sudarso, who plays Suwo, was offered a lucrative eight-year Hollywood stuntman contract and had to choose between it and Buffalo Boys; after reading the script and speaking to director Mike Wiluan, he took the film, saying his "heart and mind was in acting, not stunts anymore".
- Pevita Pearce, who plays Kiona, trained in archery for two months and worked with stuntmen from France and Thailand; she called riding the kerbau (water buffalo) a "very big challenge", describing the animals as "wild" and harder to control than horses.
- Yoshi Sudarso was the last actor to join Buffalo Boys, a month before filming began; he had moved to the United States at nine and lived there 20 years, was not fluent in Indonesian, and spent a month learning the language with help from costars Pevita Pearce and Hannah Al Rashid.
