Plot summary
Twenty-five years after the Woodsboro murders of Billy Loomis and Stu Macher, Tara Carpenter, a high school student, is attacked at home by Ghostface and hospitalized. Her estranged older sister Sam returns from Modesto with her boyfriend Richie Kirsch and rejoins Tara's friends: Wes Hicks, Liv McKenzie, Amber Freeman and the twins Chad and Mindy Meeks-Martin. Sam confides that she sees Billy Loomis in hallucinations; he had an affair with her mother, and learning he was her biological father split the family and pushed the sisters apart. Sam and Richie turn to Dewey Riley, now divorced from Gale Weathers, who warns Gale and Sidney Prescott. Mindy argues the new killer is building a requel, using Sam's bloodline to draw the original survivors back to town.

The studio got cold feet about killing off Dewey Riley and asked directors Matt Bettinelli-Olpin and Tyler Gillett to film an alternate scene showing his survival.- Kevin Williamson refused James Vanderbilt's offer to join the film because he did not want to work on a Scream project without Wes Craven; he later agreed only on the condition that the film be dedicated to Craven.
- To keep the plot from leaking before release, the filmmakers produced multiple versions of the screenplay and shot several additional scenes, a fact confirmed in April 2021.
- Scream was the first film in the series not directed by Wes Craven, who died in 2015, and the film is dedicated to him.
- Kyle Gallner auditioned for the role of Richie but was not cast in it, and was offered the part of Vince instead.
- Rian Johnson was set to make a cameo as himself, but his commitments to Glass Onion: A Knives Out Mystery prevented it, so he is only mentioned as the director of the fictional Stab 8.
