Plot summary
Kunal Chauhan is a fashion photographer and Sameer Acharya a male nurse; both are fun-loving bachelors in Miami. They cross paths at a friend's home after an evening out, then meet again while trying to rent the same flat. Neha Melwani, who works for Verve, shares that flat with her aunt, Ishita, and Ishita turns the men down because Neha wants female roommates. After an encounter with a gay soldier, Sam talks a reluctant Kunal into pretending to be his lover, and the ruse wins them the room. Meeting Neha, both regret it, since each is attracted to her. The three become friends.

- Aishwarya Rai was originally offered the lead role in Dostana but refused it because she did not want to end up with another man in her first film with her husband, Abhishek Bachchan; she later regretted the decision. Saif Ali Khan refused the role of Kunal because he was uncomfortable with a husband and wife playing the other two leads.
- Dostana was the first mainstream Indian film with a gay element and has been credited for exploring a topic largely ignored by Hindi cinema.
- Karan Johar said that after Dostana's release he received thousands of emails from children, youngsters and teenagers thanking him for making the film, because their parents were now aware of homosexuality.
- The golden swimsuit seen in Dostana was created by combining two swimsuits; because they were small, designer Manish Malhotra attached the second one to make the swimsuit seen in the film.
- Although Johar and Mansukhani wanted Chopra to wear a gown for the song "Desi Girl", designer Manish Malhotra suggested a sari, saying that if they could do a song like "Desi Girl" in Miami, Chopra could wear a sari and "treat it like a dress".
