Plot summary
A man on the run from prison arrives in Tatesville, Georgia, and presents himself as the Reverend Isaiah T. Jenkins, so the town takes him for a man of God. A fellow criminal, Yello-Curley Hinds, joins him, and the two plan to take the offerings of the congregation. Jenkins turns his attention to Isabelle Perkins, a young member of that congregation. She is in love with Sylvester, a poor young man, and he is Jenkins' long-estranged identical twin. Isabelle's mother, Martha Jane, wants her daughter to marry the reverend.

- New York's Motion Picture Commission refused Body and Soul an exhibition license, calling it immoral, sacrilegious and likely to incite to crime. Oscar Micheaux re-edited it twice, and the approved version was cut from nine reels to five, leaving his director's cut a lost film.
- Paul Robeson made his film debut in Body and Soul at the age of 27, playing the dual role of Jenkins and Sylvester. His agreement to star paid him $100 per week plus three percent of the gross after the first $40,000 in receipts.
- In 2000, Body and Soul was shown at the New York Film Festival with a new musical score written by trombonist Wycliffe Gordon and performed live by the Lincoln Center Jazz Orchestra.
