Plot summary
At the end of the Third Reich a Nazi scientist escapes into the Amazon jungle. Years later, in 1965, the adventurer John Hamilton is hired to lead investigators in search of the cause of a mysterious and incurable disease that has broken out among the natives. The trail runs up the deadly Rio del Morte toward a lost Inca city, whose wealth draws a mixed party of others, each with reasons of their own. River pirates, hostile native tribes and headhunters stand in the way. The jungle also hides the scientist, who still uses the place for experiments on people.

- River of Death was shot in South Africa during the cultural boycott, and director Steve Carver was fined by the Directors Guild of America over it. In 2020 he remained unapologetic, saying he had ignored apartheid because he was not political and did not care about it then.
- Director Steve Carver said the film was originally to be shot in Brazil, but that proved too expensive, so South Africa's Port St. Johns was chosen instead, a location he called "dangerous as all hell" and where, he said, "We were lucky no one got killed."
- Steve Carver said Herbert Lom, Robert Vaughn and Donald Pleasence carried equipment up the mountainside alongside the crew and picked up rewritten dialogue in seconds during the shoot.
