Plot summary
Dirk Green, a scientist, has built a rocketship meant to reach the Moon, and he keeps his reasons for the voyage to himself. Gary and Lon, two escaped convicts, are found hiding aboard, and Dirk forces them to serve as the crew. Steve Dayton and his fiancée June Saxton learn that the launch is close, so they go to the vessel to look it over, and they are still inside when it lifts off. The others do not know that Dirk belongs to a Lunar civilization, the last of its men, sent to Earth to bring back help for a people whose air is giving out.

- To draw teenagers, the 1958 remake of Cat-Women of the Moon added a pair of young escaped convicts, one a good kid who had made a mistake, the other an incorrigible crook, and gave them lunar love interests.
- The giant spider prop in Missile to the Moon was wire-controlled from above, and it was exactly the same "Moon spider" used five years earlier in Cat-Women of the Moon.
- A red camera filter was used to make the blue sky photograph very dark for the black-and-white film, though the result still fell far short of an ideal starry black.
- Missile to the Moon was spoofed twice on RiffTrax: first by Mystery Science Theater's Michael J. Nelson and Fred Willard, then by Nelson, Bill Corbett and Kevin Murphy, also MST3K alumni.
