Plot summary
DeWitt University's basketball team wins a championship, and the win brings an all-night scavenger hunt for the university's female students the next evening. Lynn Connors and her boyfriend Teddy Ratliff, the team's star player, celebrate at the campus diner, then join a party where freshmen trade stories about a man recently locked away in a rural sanitarium. Lynn grows jealous of Teddy's interest in Dawn Sorenson, and Mike Pryor argues with his girlfriend Sheila. Campus security officer Jim MacVey questions Mike about the fight. The next night a DJ reads the hunt's clues over the campus radio, and the girls listen on portable radios as they fan out across the grounds. The killer out that night wears the school's bear mascot costume.

- Distributor Samuel M. Sherman had the gravediggers' murder added to the film's first act, shooting it at night in Central Park, where the crew dug up a big section of ground; Sherman said he did not know whether the city had approved it.
- Filming was set to begin at Denison University in the summer of 1980, but a trustee objected to the production and the university's approval was ultimately withdrawn.
- Actress Rutanya Alda said in a 2013 interview that the producers of Girls Nite Out still owed her $5,000 for her work, money she was never paid.
- Because the film was shot on operating campuses, the filmmakers could work only over weekends, starting on a Friday and finishing on a Sunday, so the cast and crew worked in twenty-four-hour intervals.
- Producer Kevin Kurgis approached Hal Holbrook after one of the actor's Mark Twain Tonight! shows at Columbus's Mershon Auditorium and left him the screenplay; Holbrook, who had just finished John Carpenter's The Fog, accepted, and his son David also got a part.
- For the film's release in the United Kingdom and Ireland in December 1986, twenty-two seconds were excised from the original cut.
- Actress Lauren-Marie Taylor met her castmate John Didrichsen while shooting Girls Nite Out, and the couple married the year after filming finished.
