Plot summary
George Milton watches over Lennie Small, a big man with an intellectual disability, as the two take field work across California during the Great Depression. They hope to settle on a piece of land of their own rather than keep working for other people. At a ranch near Soledad, Candy, an aging one-handed ranch hand, takes them to Jackson, the ranch boss. Curley, the owner's hotheaded son, confronts them. Curley's wife Mae is bored and lonely, and she flirts with the ranch hands. George warns Lennie not to talk to her or look at her. Candy overhears the two men talk about a place of their own and offers to join them to buy the farm.

- Lon Chaney Jr., the son of silent film star Lon Chaney, had already appeared in more than 50 films when he played Lennie, but Of Mice and Men was his first major role; Burgess Meredith was a relative Hollywood newcomer cast as George.
- To satisfy the Hays Code, screenwriter Eugene Solow stripped the strong language and profanity out of John Steinbeck's story when transferring it to the screen, though director Lewis Milestone kept the piece's force.
