Plot summary
Lord Henry Blackwood, a nobleman who claims supernatural powers, has killed five women in London. Sherlock Holmes and Dr. John Watson prevent a sixth ritual murder, and Inspector Lestrade's officers take Blackwood into custody. Watson is engaged to Mary Morstan and intends to leave their rooms at 221B Baker Street, which would end the partnership. Before the hanging, Blackwood sends for Holmes and tells him that three more deaths are coming, and that Holmes cannot prevent them. Watson declares him dead after the execution. Irene Adler, an adversary who has outwitted Holmes before, then asks him to trace Luke Reordan, a man who has gone missing.

- While filming a fight scene at Chatham Dockyard in Kent in late November 2008, actor Robert Maillet, who played the henchman Dredger, accidentally punched Robert Downey Jr. in the face, bloodying him and knocking him down, though not knocking him out as first reported.
- Guy Ritchie thought Robert Downey Jr. was too old to play Sherlock Holmes, wanting a younger Holmes on a learning curve like Batman Begins, but took a chance on him; Downey had learned about the project while visiting Joel Silver's offices with his wife, producer Susan Downey.
- Robert Downey Jr. convinced Guy Ritchie to cast Rachel McAdams as Irene Adler, arguing that she would not look too young to be his love interest.
- When the production filmed at St John's Street in December, local residents complained about always having to park their cars elsewhere, so the shoot there was cut from 13 days to nine.
- Composer Hans Zimmer bought an out-of-tune piano for 200 dollars and used it throughout the scoring of Sherlock Holmes because of its quirkiness.
- Robert Downey Jr. lost weight for the part after Chris Martin, during a chat, recommended that his Sherlock Holmes look "gaunt" and "skinny".
