Plot summary
Studio executive Griffin Schraeder has agreed to a pitch meeting with screenwriter Charlie Wessler, and the outrageous sketches Charlie brings make up the movie. One sketch follows Beth, a businesswoman on a blind date with Davis, a bachelor the whole city considers a catch. Another shows Robert and Samantha giving their home-schooled son Kevin detentions, hazing and social exclusion. A grocery store intercom carries an argument between two exes to every customer in the store. A company meets about a flaw in its life-sized nude music player. Robin and Batman work a speed dating event in Gotham City while hunting a bomb threat. The rest cover a middle school date, a birthday gift for a roommate, a game of truth or dare, a 1959 basketball coach, and a woman troubled by her boyfriend's animated cat.

- George Clooney flatly refused director Peter Farrelly's pitch to play himself as a man bad at picking up women, and Colin Farrell dropped out of playing Gerard Butler's leprechaun brother, leaving Butler to do the sketch alone.
- Because it depended on when its many stars were free, the film was shot piecemeal over four years, with the crew filming for a week and then shutting down for months at a time before picking up again.
- Producer Charles Wessler struggled to book A-list actors because the shoots paid nothing beyond an $800-a-day rate for as little as two days of work, and only got stars on board through personal favors.
- The title "Movie 43" means nothing: director Peter Farrelly overheard his son talking about a film by that name, and when he discovered no such film existed, he simply took the title for his own project.
