Plot summary
Oscar and his sister Linda, Americans in Tokyo, vowed to stay together after their parents died in a car crash. Oscar sells drugs; Linda dances at a strip club. One evening, after smoking DMT, Oscar is called by his friend Victor to a bar called The Void for a deal. His friend Alex comes along and talks on the way about the Tibetan Book of the Dead and reincarnation. Inside, Oscar finds Victor distressed and the place filling with police. He locks himself in a bathroom stall and tries to flush the drugs away. Shots come through the door, and Oscar's soul lifts out of his body, looking for Linda.

- In his early twenties, while under the influence of psilocybin mushrooms, Noé saw Robert Montgomery's Lady in the Lake, a 1947 film shot entirely in a first-person perspective, and decided that if he ever made a film about the afterlife, that was how he would shoot it.
- In retrospect Noé called his earlier film Irréversible a bank robbery, a film made in order to finance Enter the Void.
- Since much of the film was set in neighborhoods known for gambling and prostitution, the producers made agreements with the Yakuza crime syndicates before filming some of the on-location scenes.
- Noé initially asked Daft Punk member Thomas Bangalter, who had composed the music for Irréversible, to create an original soundtrack for Enter the Void, but Bangalter was occupied with work on Tron: Legacy and had to decline.
- When he was already planning the film, Noé tried the psychoactive brew ayahuasca, in which the active substance is DMT, in the Peruvian jungle, where it is legal because of its traditional use as an entheogen.
- Linda was the first role to be cast: Noé found Paz de la Huerta after holding auditions in New York City, saying she had "the profile for the character because she likes screaming, crying, showing herself naked, all the qualities for it."
- Because Linda and Oscar had to be believable as siblings, Nathaniel Brown, a non-professional, was cast for his resemblance to Huerta.
- Noé had been working on different versions of the screenplay for fifteen years before the film finally went into production.
